S rastom n\u00e1dorov m\u00f4\u017eu by\u0165 spojen\u00e9 pachov\u00e9 stopy, detegovate\u013en\u00e9 niektor\u00fdmi vy\u0161\u0161\u00edmi \u017eivo\u010d\u00edchmi s citliv\u00fdm olfaktorick\u00fdm syst\u00e9mom(1). Pre klinick\u00fa prax je v\u0161ak vyu\u017eite\u013enos\u0165 t\u00fdchto zvierat z mnoh\u00fdch d\u00f4vodov neuskuto\u010dnite\u013en\u00e1. Niektor\u00e9 pr\u00e1ce nazna\u010duj\u00fa, \u017e hl\u00edstica C. elegans dok\u00e1\u017ee v\u010faka dobre vyvinut\u00fdm receptorom pre \u010duchov\u00e9 vnemy poskytn\u00fa\u0165 vhodn\u00fd syst\u00e9m pre detekciu rakoviny(2). Cie\u013eom na\u0161ej \u0161t\u00fadie bolo sledova\u0165 a vyhodnoti\u0165 chemotaktick\u00fa odpove\u010f tohto organizmu po expoz\u00edcii mo\u010du onkologick\u00fdch pacientov a kontroln\u00fdch subjektov a zisti\u0165 pr\u00edpadn\u00e9 vyu\u017eitie tejto vlastnosti pri skr\u00edningu kancerog\u00e9nnych ochoren\u00ed.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Met\u00f3dy a materi\u00e1l<\/strong><\/p>\n<p>Vzorky mo\u010du poskytli onkologick\u00ed pacienti nav\u0161tevuj\u00faci onkologick\u00e9 ambulancie v Pie\u0161\u0165anoch a Trnave; kontroln\u00e9 vzorky mo\u010du boli z\u00edskan\u00e9 od \u0161tudentov Trnavskej univerzity. Hl\u00edstice C. elegans v rovnak\u00fdch v\u00fdvojov\u00fdch \u0161t\u00e1di\u00e1ch (po synchroniz\u00e1cii) sme exponovali mo\u010du (rieden\u00e9mu a nerieden\u00e9mu) onkologick\u00fdch pacientov a kontroln\u00fdch subjektov a pre ka\u017ed\u00fa vzorku sme vypo\u010d\u00edtali chemotaktick\u00fd index (CI) pod\u013ea protokolu Margie et al., 2014(3).<\/p>\n<p>&nbsp;<\/p>\n<p><strong>V\u00fdsledky<\/strong><\/p>\n<p>\u00dadaje sme analyzovali pomocou met\u00f3dy ANOVA s vn\u00fatroskupinov\u00fdm faktorom druh vzorky (nerieden\u00e1, rieden\u00e1 vzorka) a medziskupinov\u00fdm faktorom skupina (onkologick\u00ed pacienti, kontroln\u00e1 skupina). Zistili sme signifikantn\u00fd vplyv faktoru skupina (graf 1); skupina onkologick\u00fdch pacientov mala vy\u0161\u0161ie hodnoty CI ako kontroln\u00e1 skupina: F(1;86)=196,3; p &lt; 0,0001. Faktor druh vzorky nebol signifikantn\u00fd: F(1;86) = 0,88; p = 0,350, ani interakcia faktorov skupina x druh vzorky nebola signifikantn\u00e1: F(1;86) = 1,43; p = 0,234. Pomocou logistickej bin\u00e1rnej regresie sme zistili, \u017ee hodnoty CI dok\u00e1\u017eu efekt\u00edvne rozl\u00ed\u0161i\u0165 onkologick\u00fdch pacientov od os\u00f4b z kontrolnej skupiny (85,2 % spr\u00e1vne identifikovan\u00fdch pacientov pod\u013ea CI nerieden\u00fdch vzoriek; 84,1 % spr\u00e1vne identifikovan\u00fdch pacientov pod\u013ea CI rieden\u00fdch vzoriek).<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Diskusia<\/strong><\/p>\n<p>Onkologick\u00e9 ochorenia s\u00fa jednoduch\u0161ie lie\u010dite\u013en\u00e9 v po\u010diato\u010dnom \u0161t\u00e1diu, v ktorom sa sympt\u00f3my e\u0161te neprejavuj\u00fa. Hirotsu et al. (2015) vo svojej pr\u00e1ci pouk\u00e1zal na mo\u017enos\u0165 vyu\u017ei\u0165 na v\u010dasn\u00e9 odhalenie rakoviny chemotaktick\u00e9 testy s pou\u017eit\u00edm nemat\u00f3d. Autori \u0161t\u00fadie v teste porovnali \u201ewild type\u201c C. elegans a C. elegans s genetickou abl\u00e1ciou neur\u00f3nov zodpovedn\u00fdch za \u010duchov\u00e9 vnemy. Upraven\u00e9 nemat\u00f3dy s oslaben\u00fdmi olfaktorick\u00fdmi schopnos\u0165ami na rozdiel od \u201ewild type\u201c u\u017e nevykazovali pozit\u00edvnu chemotaxiu k mo\u010du onkologick\u00fdch pacientov(2). Chemotaxia by teda pravdepodobne mohla by\u0165 spojen\u00e1 s rozpozn\u00e1van\u00edm \u0161pecifick\u00fdch pachov\u00fdch st\u00f4p t\u00fdmito nemat\u00f3dami. Toto je v s\u00falade s na\u0161imi v\u00fdsledkami, ktor\u00e9 nazna\u010duj\u00fa, \u017ee C. elegans dok\u00e1\u017eu odhali\u0165 ur\u010dit\u00e9 stopy z\u00e1pachu v mo\u010di onkologick\u00fdch pacientov bez oh\u013eadu na typ rakoviny a riedenie vzoriek mo\u010du.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Z\u00e1ver<\/strong><\/p>\n<p>Na z\u00e1klade na\u0161ich v\u00fdsledkov predpoklad\u00e1me, \u017ee mo\u010d onkologick\u00fdch pacientov obsahuje \u0161pecifick\u00e9 pachov\u00e9 stopy, ktor\u00e9 sa prejavili v chemotaktick\u00fdch testoch s pou\u017eit\u00edm C. elegans. Do bud\u00facnosti by v\u0161ak boli potrebn\u00e9 \u010fal\u0161ie testy, ktor\u00e9 by pomohli pomenova\u0165 l\u00e1tky v mo\u010di t\u00fdchto pacientov, zodpovedaj\u00face za chemotaktick\u00fa odpove\u010f hl\u00edstic a objasni\u0165 \u010fal\u0161ie s\u00favislosti spojen\u00e9 s n\u00e1stupom rakoviny.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>LITERAT\u00daRA<\/strong><br \/>\n1. Larvae of C. elegans were exposed to samples of urine of oncologic patients and control subjects. We calculated the chemotactic indices for each sample. The statistical analysis revealed that urine samples in the group of oncologic patients had significantly higher CI values than in the control group (p &lt; 0.0001).
