Ich trofozoity dok\u00e1\u017eu vplyvom nepriazniv\u00fdch podmienok encystova\u0165(1). Akantam\u00e9by s\u00fa p\u00f4vodcami z\u00e1va\u017en\u00fdch infekci\u00ed \u010dloveka (granulomat\u00f3zna am\u00e9bov\u00e1 encefalit\u00edda, akantam\u00e9bov\u00e1 keratit\u00edda, ko\u017en\u00e9 a diseminovan\u00e9 infekcie)(2), na ktor\u00e9 doposia\u013e neexistuje jednotn\u00e1 a spo\u013eahliv\u00e1 terapia. To je d\u00f4vodom pre h\u013eadanie nov\u00fdch potenci\u00e1lnych terapeut\u00edk. Am\u00e9bic\u00eddny efekt metronidazolu sa \u00faspe\u0161ne vyu\u017e\u00edva v lie\u010dbe ameb\u00f3zy sp\u00f4sobenej druhom Entamoeba histolytica(3). Aplik\u00e1cia v pr\u00edpade akantam\u00e9bov\u00fdch infekci\u00ed nepreuk\u00e1zala dostato\u010dn\u00fd terapeuticky \u00fa\u010dinok(4). V\u00fdskumy sa s\u00fastre\u010fuj\u00fa aj na testovanie cytotoxicity fosf\u00f3niov\u00fdch sol\u00ed. Ned\u00e1vne \u0161t\u00fadie potvrdili ich am\u00e9bic\u00eddny \u00fa\u010dinok aj na kme\u0148och akantam\u00e9b(5). Cie\u013eom v\u00fdskumu bolo definovanie cytotoxicity nov\u00e9ho metronidazolov\u00e9ho deriv\u00e1tu (fosf\u00f3niovej soli) na dvoch izol\u00e1toch rodu Acanthamoeba. Nako\u013eko je testovan\u00fd deriv\u00e1t novosyntetizovanou zl\u00fa\u010deninou, jeho cystic\u00eddny \u00fa\u010dinok bude predmetom \u010fal\u0161ieho sk\u00famania.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Materi\u00e1l a metodika<\/strong><\/p>\n<p>Experiment\u00e1lne bol testovan\u00fd cytotoxick\u00fd \u00fa\u010dinok deriv\u00e1tu metronidazolu \u2013 fosf\u00f3niovej soli (FS) a metronidazolu na dvoch klinick\u00fdch izol\u00e1toch: A. lugdunensis (AcaVNAK02), A. quina (AcaVNAK03). Na axenick\u00fdch kult\u00farach trofozoitov bolo celkovo testovan\u00fdch 6 koncentr\u00e1ci\u00ed zl\u00fa\u010den\u00edn: 500 \u03bcl; 250 \u03bcl; 125 \u03bcl; 62,5 \u03bcl; 31,25 \u03bcl a 15,6 \u03bcl, pri teplote 37 \u00b0C v steriln\u00fdch podmienkach. V\u00fdsledky boli od\u010d\u00edtan\u00e9 po 1, 24 a 48 hodin\u00e1ch. Stanoven\u00e1 bola minim\u00e1lna trofoc\u00eddna koncentr\u00e1cia (MTC) \u2013 najni\u017e\u0161ia koncentr\u00e1cia zl\u00fa\u010deniny so 100 % trofoc\u00eddnym \u00fa\u010dinkom.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>V\u00fdsledky<\/strong><\/p>\n<p>V priebehu experimentu v\u0161etky koncentr\u00e1cie FS prejavili inhibi\u010dn\u00fd efekt (obr\u00e1zok 1) na oba kmene akantam\u00e9b. Pri kmeni A. lugdunensis boli hodnoty MTC po 1 aj 24 hodin\u00e1ch\u00a0 na rovnakej \u00farovni 500 \u03bcM, vy\u0161\u0161ia cytotoxicita zl\u00fa\u010deniny sa prejavila a\u017e po 48 hodin\u00e1ch (MTC 250 \u03bcM) (tabu\u013eka 1). Kme\u0148 A. quina prejavil vy\u0161\u0161iu citlivos\u0165, s hodnotami MTC 500 \u03bcM po 1 hodine a 250 \u03bcM po 24 aj 48 hodin\u00e1ch. Z\u00edskan\u00e9 hodnoty MTC potvrdili vy\u0161\u0161iu \u00fa\u010dinnos\u0165 metronidazolov\u00e9ho deriv\u00e1tu v porovnan\u00ed s referen\u010dn\u00fdm metronidazolom. MTC metronidazolu dosahovalo pri oboch kme\u0148och rovnak\u00e9, vysok\u00e9 hodnoty &gt; 500 \u03bcM vo v\u0161etk\u00fdch troch \u010dasov\u00fdch intervaloch.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Diskusia<\/strong><\/p>\n<p>Testovan\u00fd metronidazolov\u00fd deriv\u00e1t \u2013 fosf\u00f3niov\u00e1 so\u013e, preuk\u00e1zal v\u00fdrazne v\u00e4\u010d\u0161\u00ed cytotoxick\u00fd \u00fa\u010dinok v porovnan\u00ed s referen\u010dn\u00fdm metronidazolom. Amebic\u00eddny \u00fa\u010dinok na rovnak\u00fdch kme\u0148och akantam\u00e9b preuk\u00e1zali ned\u00e1vno aj fosf\u00f3niov\u00e9 soli kyseliny rozmar\u00ednovej(5). Tieto zistenia nazna\u010duj\u00fa potrebu \u010fal\u0161ieho sk\u00famania metronidazolu ako v\u00fdchodiskovej zl\u00fa\u010deniny pre synt\u00e9zu nov\u00fdch potenci\u00e1lne am\u00e9bic\u00eddnych zl\u00fa\u010den\u00edn, ako aj nutnos\u0165 \u010fal\u0161ieho testovania am\u00e9bic\u00eddneho \u00fa\u010dinku fosf\u00f3niov\u00fdch sol\u00ed.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Z\u00e1ver<\/strong><\/p>\n<p>Zo z\u00edskan\u00fdch v\u00fdsledkov mo\u017eno potvrdi\u0165 vy\u0161\u0161iu \u00fa\u010dinnos\u0165 novosyntetizovan\u00e9ho deriv\u00e1tu oproti metronidazolu. Na z\u00e1klade v\u00fdsledkov vypl\u00fdva potreba \u010fal\u0161ieho sk\u00famania dan\u00e9ho deriv\u00e1tu na in\u00fdch kme\u0148och akantam\u00e9b.<\/p>\n<p><strong>Po\u010fakovanie:<\/strong> Pr\u00e1ca vznikla s podporou grantov VEGA 1\/0389\/19, VEGA 1\/0054\/19, KEGA 013TTU-4\/2019 a UK\/173\/2020. The cytotoxic effect of a new phosphonium salt, a metronidazole derivate, was investigated on two clinical isolates of species Acanthamoeba lugdunensis and Acanthamoeba quina. The minimal trophocidal concentrations \u2013 MTC was measured during three different time intervals. The phosphonium salt was significantly more effective than metronidazole.
